- CRISPR/CAS9이나 siRNA/shRNA 등을 이용해서 KO/KD 모델을 만든 뒤, protein expression 여부를 비교

- in situ hybridization, FISH, quantitative PCR, RNA-seq, mass spectrometry 등 antibody를 사용하지 않는 실험 결과와 antibody를 사용하는 실험 결과를 비교

Antibody vs. Antibody

- 하나의 traget을 잡는 서로 다른 antibody들을 이용해서 하나의 동일한 실험을 진행

- 모든 antibody에서의 결과가 동일하다면 antibody specificity가 좋다는 것을 확인할 수 있음.

Tag Ab vs. Target Ab

- Tagging이 붙어있는 protein을 이용해서 실험 진행

- Tag antibody와 Target antibody 결과 비교함으로써 antibody specificity 확인

​

Treated vs. Untreated

- 잘 알려진 생물학적, 화학적 reagent를 이용해서 protein expression을 조절한 뒤 실험 진행

- Protein expression이 변하는 것을 확인함으로써 antibody specificity 확인!
